Your boss and uncle did that to you because they knew you wouldn't respond aggressively. I used to deal with a large number of people that thought treating me with respect was optional. What your boss and uncle did was disrespectful and they did it because, in short, they do not respect you and are not intimidated by you.
Trust, if you had a rep of being a finger rolling, neck wagging, cuss a nicca out type of chick, they wouldn't have dared to do such a thing to you. And it is really sad and pathetic that one has to act like the aforementioned in order to be treated with respect by some people (and it is not even authentic, genuine "respect" it is fear and intimidation; folks like that leave the "she beasts" alone and go after the people they view as "weak").
It doesn't matter if people know or don't know that your hair is a weave/wig/phony pony whatever. People like that pick and choose who they treat with respect and dignity.
You need to let your inner b!tch out more often and let these people know what type of treatment you will and will not accept. I've managed to significantly reduce that "large number of people who think respecting me is optional" that I mentioned at the onset of my post. These people weren't family or co-workers though which admittedly does make your situation more precarious.
At the bare minimum I would suggest you address your boss as you addressed your uncle (informing him that you felt disrespected and violated and that you behave in a professional manner and want to likewise be treated in a professional manner). It is not about "not being able to take a joke" it is about respect and people treating you the way they themselves would want to be treated.
